I'd rather not keep paying a monthly subscription just to use the Script Dept (script supervisor) features. Can I buy MovieSlate Pro once instead and still get Script Dept?

Script Dept (the script supervisor workflow) is only available as a monthly subscription — there is no one-time purchase option for it. The good news is that the Script Dept subscription includes the Pro Features bundle, so you don't need to buy or subscribe to Pro Features separately while you have Script Dept.
Pro Features on their own are offered either as a monthly subscription or as a one-time purchase — but a Pro Features purchase does not include the script supervisor (Script Dept) workflow.

One tip: you should be subscribed to either Script Dept or Pro Features, not both. Since Script Dept already includes Pro Features, paying for both is redundant — you can cancel the duplicate in the App Store under your account's Subscriptions.
